The surface-enhanced Raman scattering (SERS) technique provides outstanding molecular fingerprint identification and high sensitivity of analytes. Herein, colloidal sphere-shaped gold nanoparticles (Au-NPs) trapped in concentric rings of the Bessel beam generated from the optical fiber-based negative axicon has been reported as a SERS substrate. With the trapping of Au-NPs, the SERS ability of colloidal Au-NPs improved, and the average enhancement factor (AEF) of the rhodamine-6G (R6G) and 4-aminothiophenol (4-ATP) molecules can reach up to the order of 107. Control experiments were also carried out with the trapping of Au-NPs by Gaussian beam illumination, without any illumination of the light and with the trapping of Au-NPs by the Bessel beam illumination on a silver (Ag)-coated silicon (Si) substrate with a self-assembled monolayer (SAM) of 4-ATP. Theoretical studies were also carried out using the finite element method (FEM) to identify the hotspots generated in the gaps formed between the Au-NPs, leading to an enhancement in the SERS signal of the molecules, and the results were consistent with the experimentally determined AEFs. The obtained results demonstrate that the proposed SERS technique is stable. Machine learning (ML) is effective to establish the complicated trilateral relationship among structures, properties, and photovoltaic performance, which is fundamental issue in developing novel materials for improving power conversion efficiency (PCE) of organic solar cells (OSCs). Herein, we constructed the database of 397 donor–acceptor pairs of OSCs with photovoltaic parameters and descriptor sets, which include donor–acceptor weight ratio within the active layer of the OSCs, root mean square of roughness, and 1024-bit Morgan molecular fingerprint for donor (Fp-D) and acceptor (Fp-A). The ML models random forest (RF), adaptive boosting (AdaBoost), extra trees regression, and gradient boosting regression trees were trained based on the descriptor set. The metrics determination coefficient (R2), Pearson correlation coefficient (r), root mean square error, and mean absolute error were selected to evaluate ML model performances. The results showed that the RF model exhibits the highest accuracy and stability for PCE prediction among these four ML models. Moreover, based on the decomposition of non-fullerene acceptors L8-BO, BTP-ec9, AQx-2, and IEICO, 20 acceptor molecules with symmetric A–D–A and A–π–D–π–A architectures were designed. The photovoltaic parameters of the designed acceptors were predicted using the trained RF model, and the virtual screening of designed acceptors was conducted based on the predicted PCE. The results indicate that six designed acceptors can reach the predicted PCE higher than 12% when P3HT was adopted as a donor. Wine cultivars are available to growers in multiple clonal selections with agronomic and enological differences. Phenotypic differences between clones originated from somatic mutations that accrued over thousands of asexual propagation cycles. Genetic diversity between grape cultivars remains unexplored, and tools to discriminate unequivocally clones have been lacking. This study aimed to uncover genetic variations among a group of clonal selections of 4 important Vitis vinifera cultivars: Cabernet sauvignon, Sauvignon blanc, Chardonnay, and Merlot, and use this information to develop genetic markers to discriminate the clones of these cultivars. We sequenced with short-read sequencing technology the genomes of 18 clones, including biological replicates for a total of 46 genomes. Sequences were aligned to their respective cultivars reference genome for variant calling. We used reference genomes of Cabernet sauvignon, Chardonnay, and Merlot and developed a de novo genome assembly of Sauvignon blanc using long-read sequencing. On average, 4 million variants were detected for each clone, with 74.2% being single nucleotide variants and 25.8% being small insertions or deletions (InDel). The frequency of these variants was consistent across all clones. From these variants, we validated 46 clonal markers using high-throughput amplicon sequencing for 77.7% of the evaluated clones, most of them small InDel. These results represent an advance in grapevine genotyping strategies and will benefit the viticulture industry for the characterization and identification of the plant material.

We present the first experimental application of coherent Raman spectroscopy (CRS) on the ro-vibrational ν2 mode spectrum of methane (CH4). Ultrabroadband femtosecond/picosecond (fs/ps) CRS is performed in the molecular fingerprint region from 1100 to 2000 cm−1, employing fs laser-induced filamentation as the supercontinuum generation mechanism to provide the ultrabroadband excitation pulses. We introduce a time-domain model of the CH4ν2 CRS spectrum, including all five ro-vibrational branches allowed by the selection rules Δv = 1, ΔJ = 0, ±1, ±2; the model includes collisional linewidths, computed according to a modified exponential gap scaling law and validated experimentally. The use of ultrabroadband CRS for in situ monitoring of the CH4 chemistry is demonstrated in a laboratory CH4/air diffusion flame: CRS measurements in the fingerprint region, performed across the laminar flame front, allow the simultaneous detection of molecular oxygen (O2), carbon dioxide (CO2), and molecular hydrogen (H2), along with CH4. Fundamental physicochemical processes, such as H2 production via CH4 pyrolysis, are observed through the Raman spectra of these chemical species. In addition, we demonstrate ro-vibrational CH4v2 CRS thermometry, and we validate it against CO2 CRS measurements. Bone samples continue to be a challenging source of DNA, requiring tailored DNA extraction methods to maximize DNA profile quality. Several modified methods were compared to the current bone method based on updates in the literature. Results showed that increasing the EDTA pH, adding a surfactant and using an alternative DNA extraction buffer were not advantageous over the full demineralization method currently implemented in the laboratory. However, as a result of the modification study, it was observed that shorter incubation periods improved both the quantity and quality of DNA profiles obtained from bones. [ABSTRACT FROM AUTHOR]

Comparisons of DNA profiles recovered from the crime-scene and a person through short tandem repeat (STR) analysis using capillary electrophoresis (CE) is the gold standard method of determining a match. A comparison of STR profiles generated using CE to massively parallel sequencing (MPS) methods was undertaken. The results showed CE methods recovered high percentages of autosomal or Y-STR markers individually, however, MPS produced more information at low inputs and a higher number of STR markers overall, showing the potential of MPS to provide additional data to assist in solving major crimes. Face touching is a common habit that can have implications on the outcome of criminal investigations. When conducting activity level assessments, it is important to be aware of any factors that may impact the type of profile obtained from the sample of interest. This may include the number, duration, location, and manner of contacts that a person makes with themselves. In this study we have recorded the contacts made by different individuals, of their hands with their face, over a 1 hr period, while working within an office. All participants were observed to regularly touch their face. Swabs of the face of some participants were subsequently collected for DNA profiling. This study proposed a new PCF design to extend the SC generation beyond the long wavelength edge of the MIR molecular fingerprint region. The proposed design was made of AsSe2 glass as a background material whose has a significant nonlinear refractive index at 11 × 10−18 m2 W−1. The proposed PCF design was consist of five layers of circular air holes arranged in a triangular lattice of pitch size (Λ = 2 µm). The air hole diameters of the first layer were different in size with other layers to give more flexibility in the control of the chromatic dispersion. The geometrical parameters of the PCF design are modified to achieve the anomalous dispersion regime. We applied the Finite Difference Method to compute the linear and nonlinear parameters of the proposed PCF structure including dispersion, loss, effective mode area and the Kerr nonlinearity. The Nonlinear Schrödinger Equation is numerically solved by using Split-Step Fourier method to evaluate MIR SC generation. The simulated results showed that by using only 4 cm of the suggested PCF design pumped with 3 kW peak power and 330 fs pulses at 8 μm wavelength in the anomalous dispersion regime is enough to generate coherent MIR SC covering from 5 to 21 μm. Molecular identification of extremely compromised human remains in forensic field is usually performed from DNA typing of bones, which are a difficult sample to work with. Moreover, autosomal STR profiles do not always result in the identification of the donor due to lack of comparisons or non-hit throughout database searching. An attempt to overcome these issues is represented by fingernails as an alternative DNA source and Y-STRs typing to infer both geographical and familial ancestry of the unknown donor. In this study, we analyzed both 24 autosomal and 27 Y-chromosome STRs from unidentified human remains (UHRs) of five males recovered from the water near the southwestern coast of Sardinia by the Italian Harbor Master's Office. Nail clippings provided an optimal source of autologous DNA for molecular identification in a very short time, producing complete autosomal and Y-STR profiles even under conditions of high body degradation. Unfortunately, no match neither compatibility occurred using autosomal STRs (aSTRs), initially. Upon analyzing the Y-haplotypes, we found out they had already been observed in northern Africa, providing us important investigative leads. This prompted the International Criminal Police Organization (INTERPOL) to provide us with references of alleged relatives that were then confirmed to be related. The use of fingernails represents an excellent DNA source especially for genetic identification of decomposed bodies recovered in seawater environment. Notably, DNA extracted from nails gave high-quality Y-STR haplotypes by which predicting paternal ancestry of the unidentified donors may result fundamental in the forensic investigative context. Studying DNA transfer and persistence has become increasingly important over the last decade, due to the impressive sensitivity of modern DNA detection methods in forensic genetics. To improve our understanding of background DNA that could also potentially be transferred, we analyzed the DNA composition on the outside of sleeve cuffs and sampled DNA directly from the hands of four different collaborators upon their arrival at work during 25 working days. Sampling of their hands was repeated after several hours working in our department. The shedder status of the participants, as assumed from previous internal studies, was well re-produced in the study. However, we noticed that the DNA shedding capacity could also change drastically during the day, with one participant showing a more than sixfold increase between hands sampled in the morning and hands sampled in the afternoon. As expected, poor DNA shedders carry more relative amounts of non-self-DNA on their hands than good shedders. Non-self-alleles were detected in 95% of the samples. We also observed potential effects of hand washing and the mode of transport to get to work on the DNA amount. People living with family members occasionally carried their DNA on their hands and more frequently on their sleeve cuffs. Sleeve cuffs, as being close to our hands, have a large potential to transfer DNA from one place to another, yet they have sparsely been studied as DNA transfer intermediates so far. In general, we collected consistently more DNA from the sleeve cuffs than from the hands of the participants, demonstrating their importance as potential transfer vectors. More DNA was recovered from sleeve cuffs made of synthetic fabric than from cuffs made of cotton or leather. In the afternoon, DNA from co-habitant family members could not be detected on the hands anymore and the detection of profiles from colleagues became more frequent. From two out of 100 analyzed sleeve cuffs and two out of 200 sampled hands, we established unknown major DNA profiles that would have been suitable for an entry in the national DNA database. In a forensic scenario, if biological stains are found in very small quantities, these are usually collected for DNA analyses, considered essential for the forensic investigation and thus excluding possible investigations by other forensic disciplines as forensic toxicology. We developed an experimental study to evaluate the feasibility of analyzing DNA extraction residues obtained from DNA extraction procedures to perform toxicological analysis, with the aim to extract both genetic and toxicological information without affecting or compromising the genetic sample and/or DNA extraction. DNA extraction from four blood samples (fortified with 5 molecules of interest with a final concentrations of 1 µg/mL, 100 ng/mL, 10 ng/mL and 5 ng/mL, respectively) were analyzed with QIAGEN QIAmp® DNA Mini kit. Three waste residues collected from the DNA extraction were analyzed for the toxicological investigation via Solid-Phase Extraction and High-Performance Liquid Chromatography—Tandem Mass Spectrometry analyses (Thermo Scientific™ TSQ Fortis™ II Triple-Quadrupole Mass Spectrometer). The analytical investigation revealed that our analytes of interest were detected in two different residues of the DNA extraction procedure, allowing both genetic and toxicological analyses without affecting the DNA identification. The mid‐infrared (mid‐IR) region is attractive for spectroscopy because this range has molecular fingerprint information. Existing FTIR‐based systems are limited by their complex optical systems, highlighting the ongoing necessity to advance miniaturization and simplify these systems. Here, a compact mid‐IR spectroscopy is introduced in the mid‐IR range, utilizing a continuously variable mid‐IR filter. This filter is made from Germanium (Ge) and BaF2 and is produced using physical vapor deposition. Continuously varying wavelengths of mid‐IR light are transmitted from different areas of the filter. These different wavelengths of light enter each pixel of the microbolometer array. Calibration is conducted using a sample material to match each pixel of the microbolometer array to its corresponding wavelength. Additionally, a deconvolution algorithm is applied to sharpen the acquired spectrum. This results in acquisition of high signal‐to‐noise ratio mid‐IR spectra in the LWIR region. Using this technology, mid‐IR spectroscopy for several types of samples like polymer film, and gas molecules is possible. Diseases related to the central nervous system (CNS) are major health concerns and have serious social and economic impacts. Developing new drugs for CNS-related disorders presents a major challenge as it actively involves delivering drugs into the CNS. Therefore, it is imperative to develop in silico methodologies to reliably identify potential lead compounds that can penetrate the blood–brain barrier (BBB) and help to thoroughly understand the role of different physicochemical properties fundamental to the BBB permeation of molecules. In this study, we have analysed the chemical space of the CNS drugs and compared it to the non-CNS-approved drugs. Additionally, we have collected a feature selection dataset from Muehlbacher et al. (J Comput Aided Mol Des 25(12):1095–1106, 2011. 10.1007/s10822-011-9478-1) and an in-house dataset. This information was utilised to design a molecular fingerprint that was used to train machine learning (ML) models. The best-performing models reported in this study achieved accuracies of 0.997 and 0.98, sensitivities of 1.0 and 0.992, specificities of 0.971 and 0.962, MCCs of 0.984 and 0.958, and ROC-AUCs of 0.997 and 0.999 on an imbalanced and a balanced dataset, respectively. They demonstrated overall good accuracies and sensitivities in the blind validation dataset. The reported models can be applied for fast and early screening drug-like molecules with BBB potential. Furthermore, the bbbPythoN package can be used by the research community to both produce the BBB-specific molecular fingerprints and employ the models mentioned earlier for BBB-permeability prediction. The brown bear (Ursus arctos L., 1758) is a widespread bear species inhabiting the forest zone of Eurasia, including the Republic of Yakutia. The association with forest habitats explains why the Pleistocene findings of U. arctos fossils are rare in the northern part of Eastern Siberia, where open steppe-tundra and steppefied landscapes prevailed during the Pleistocene. Fossils of U. arctos that have been found on the territory of Yakutia are dated since the beginning of the Middle Pleistocene. These are mainly the skulls and bones of the postcranial skeleton. In the present study, using comparative morphological analysis, computed tomography, and DNA sequencing, we describe a first mummified carcass of a brown bear individual that inhabited the New Siberian Islands (Northeast Siberia) in the Middle Holocene, approximately 3,500 years BP, which was found in the permafrost of Bolshoy Lyakhovsky Island, Russia, in 2020. Biological membranes have a stunning ability to adapt their composition in response to physiological stress and metabolic challenges. Little is known how such perturbations affect individual organelles in eukaryotic cells. Pioneering work has provided insights into the subcellular distribution of lipids in the yeast Saccharomyces cerevisiae, but the composition of the endoplasmic reticulum (ER) membrane, which also crucially regulates lipid metabolism and the unfolded protein response, remains insufficiently characterized. Here, we describe a method for purifying organelle membranes from yeast, MemPrep. We demonstrate the purity of our ER membrane preparations by proteomics, and document the general utility of MemPrep by isolating vacuolar membranes. Quantitative lipidomics establishes the lipid composition of the ER and the vacuolar membrane. Our findings provide a baseline for studying membrane protein biogenesis and have important implications for understanding the role of lipids in regulating the unfolded protein response (UPR). The combined preparative and analytical MemPrep approach uncovers dynamic remodeling of ER membranes in stressed cells and establishes distinct molecular fingerprints of lipid bilayer stress. Synopsis: This study describes a new technology for isolating organelle membranes from yeast, MemPrep. Combined with quantitative lipidomics, MemPrep establishes molecular fingerprints of membrane-based forms of endoplasmic reticulum (ER) stress. MemPrep provides quantitative insight into the compositions of the yeast vacuole and ER membrane. The ER membrane composition is significantly distinct from the whole cell lipidome. Lipid metabolic perturbation causes dramatic changes of the ER membrane composition, which sometimes trigger ER stress. Inositol depletion and prolonged proteotoxic stress lead to distinct lipid fingerprints and membrane proteomes. Increased membrane thickness and low levels of anionic lipids are common denominators of lipid bilayer stress. Combined with proteomics and quantitative lipidomics, this method for purification of specific yeast membranes establishes molecular fingerprints of membrane-based forms of endoplasmic reticulum (ER) stress. In view of the current biodiversity crisis and our need to preserve and improve ecosystem functioning, efficient means for characterizing and monitoring biodiversity are required. DNA barcoding, especially when coupled with new sequencing technologies, is a promising method that can, in principle, also be employed by taxonomic lay people. In this study we compare the performance of DNA barcoding by means of a third-generation sequencing technology, nanopore sequencing with classical Sanger sequencing, based on a sample of invertebrates collected from moss pads in a bog in Austria. We find that our nanopore sequencing pipeline generates DNA barcodes that are at least as good as barcodes generated with Sanger sequencing, with the MinION producing better results than the Flongle flowcell. We further find that while many arthropod taxa are well covered in the international reference DNA barcode database BOLD, this clearly is not the case for important taxa like mites and springtails, which hampers large-scale biodiversity assessments. Based on examples from our study we further highlight which factors might be responsible for ambiguous species identification based on BOLD and how this can, at least partly, be solved. The polymerase chain reaction (PCR) has played a fundamental role in our understanding of the world, and has applications across a broad range of disciplines. The introduction of PCR into forensic science marked the beginning of a new era of DNA profiling. This era has pushed PCR to its limits and allowed genetic data to be generated from trace DNA. Trace samples contain very small amounts of degraded DNA associated with inhibitory compounds and ions. Despite significant development in the PCR process since it was first introduced, the challenges of profiling inhibited and degraded samples remain. This review examines the evolution of the PCR from its inception in the 1980s, through to its current application in forensic science. The driving factors behind PCR evolution for DNA profiling are discussed along with a critical comparison of cycling conditions used in commercial PCR kits. Newer PCR methods that are currently used in forensic practice and beyond are examined, and possible future directions of PCR for DNA profiling are evaluated. [ABSTRACT FROM AUTHOR]

Immortelle (Helichrysum italicum) essential oil is one of the most valuable essential oils with its commercial importance and favorable characteristic properties in various cosmetic products and food products. Current research performed a comprehensive characterization of Immortelle essential oil using powerful and robust analytical techniques. The relative volatile composition of the Immortelle essential oil from Turkey was determined by GC-MS (gas chromatography-mass spectrometry) technique. Thirty-eight volatile compounds were identified. Average total phenolic content and DPPH radical scavenging activities (IC50) were determined as 54.24 mg GAE/mL essential oil and 0.27 mg/ml, respectively. FTIR and Raman spectroscopy fingerprints were determined for chemical characterization. The thermal profile was obtained by DSC (differential scanning calorimetry) analysis. The Immortelle essential oil (12.5 μg/mL) inhibited the growth of Caco 2 adenocarcinoma cells at the ratio of 48% without any deterioration in healthy L929 fibroblast cells according to the cytotoxicity analyses. Unlike chronological age, biological age is a strong indicator of health of an individual. However, the molecular fingerprint associated with biological age is ill‐defined. To define a high‐resolution signature of biological age, we analyzed metabolome, circulating senescence‐associated secretome (SASP)/inflammation markers and the interaction between them, from a cohort of healthy and rapid agers. The balance between two fatty acid oxidation mechanisms, β‐oxidation and ω‐oxidation, associated with the extent of functional aging. Furthermore, a panel of 25 metabolites, Healthy Aging Metabolic (HAM) index, predicted healthy agers regardless of gender and race. HAM index was also validated in an independent cohort. Causal inference with machine learning implied three metabolites, β‐cryptoxanthin, prolylhydroxyproline, and eicosenoylcarnitine as putative drivers of biological aging. Multiple SASP markers were also elevated in rapid agers. Together, our findings reveal that a network of metabolic pathways underlie biological aging, and the HAM index could serve as a predictor of phenotypic aging in humans. [ABSTRACT FROM AUTHOR]

The aggregation-induced emission (AIE) effect exhibits a significant influence on the development of luminescent materials and has made remarkable progress over the past decades. The advancement of high-performance AIE materials requires fast and accurate predictions of their photophysical properties, which is impeded by the inherent limitations of quantum chemical calculations. In this work, we present an accurate machine learning approach for the fast predictions of quantum yields and wavelengths to screen out AIE molecules. A database of about 563 organic luminescent molecules with quantum yields and wavelengths in the monomeric/aggregated states was established. Individual/combined molecular fingerprints were selected and compared elaborately to attain appropriate molecular descriptors. Different machine learning algorithms combined with favorable molecular fingerprints were further screened to achieve more accurate prediction models. The simulation results indicate that combined molecular fingerprints yield more accurate predictions in the aggregated states, and random forest and gradient boosting regression algorithms show the best predictions in quantum yields and wavelengths, respectively. Given the successful applications of machine learning in quantum yields and wavelengths, it is reasonable to anticipate that machine learning can serve as a complementary strategy to traditional experimental/theoretical methods in the investigation of aggregation-induced luminescent molecules to facilitate the discovery of luminescent materials. Okra, Abelmoschus esculentus (L.) Moench, being highly susceptible to yellow vein mosaic virus (YVMV) disease warrants its genetic improvement for resistance. Applied mutagenesis programme using two optimum doses of gamma radiation, namely, 350 Gy and 450 Gy radiation, was administered for the variety Pusa Sawani rated as excellent for fruit quality but highly susceptible to YVMV disease. Two selected putative mutant families, namely, 350//10///3‐9////28 and 450//66///2‐4////39, isolated in the M5 generation with slightly different plant morphology as compared to the parental genotype, Pusa Sawani, showed consistent resistance against YVMV disease. Upon evaluation in the M6 generation, eight morphological characteristics and five quantitative characteristics differed significantly among the mutants and Pusa Sawani. Proximate compositions and enzyme activity in leaf were significantly higher in the two YVMV‐resistant mutants. Disease screening under artificial inoculation in the insect proof cages confirmed YVMV resistance in these mutants. DNA fingerprinting further validated the alterations occurred in these two isolated mutants compared to the parental genotype. Harmful alcohol consumption is a major socioeconomic burden to the health system, as it can be the cause of mortality of heavy alcohol drinkers. The dopaminergic (DAergic) system is thought to play an important role in the pathogenesis of alcohol drinking behaviour; however, its exact role remains elusive. Fibroblast growth factor 2 (FGF‐2), a neurotrophic factor, associated with both the DAergic system and alcohol consumption, may play an important role in DAergic neuroadaptations during alcohol abuse. Within this study, we aimed to clarify the role of endogenous FGF‐2 on the DAergic system and whether there is a possible link to alcohol consumption. We found that lack of FGF‐2 reduces the alcohol intake of mice. Transcriptome analysis of DAergic neurons revealed that FGF‐2 knockout (FGF‐2 KO) shifts the molecular fingerprint of midbrain dopaminergic (mDA) neurons to DA subtypes of the ventral tegmental area (VTA). In line with this, proteomic changes predominantly appear also in the VTA. Interestingly, these changes led to an altered regulation of the FGF‐2 signalling cascades and DAergic pathways in a region‐specific manner, which was only marginally affected by voluntary alcohol consumption. Thus, lack of FGF‐2 not only affects the gene expression but also the proteome of specific brain regions of mDA neurons. Bermudagrass (Cynodon spp.) is a drought-resistant warm-season turfgrass adapted to the southern and transitional zones in the United States. Multiple hybrid cultivars have been developed and released for use as turfgrass, and others are still undergoing development. Increasing genetic diversity of commercial cultivars is vital to stress tolerance. A DNA profiling study of 21 experimental selections from the Oklahoma State University turfgrass breeding program and 11 cultivars was conducted using 51 simple sequence repeat primer pairs across the bermudagrass genome. A pairwise genetic relationship analysis of the genotypes using 352 polymorphic bands showed genetic similarity coefficients ranging from 0.59 to 0.89. The average pairwise population differentiation values were 0.012 for the 11 cultivars and 0.169 for the 21 selections. A cluster analysis using the unweighted paired group with the arithmetic average method grouped the entries into six clusters. A correlation analysis identified different levels of pairwise genetic relationships among the entries that largely reflected parental relationship. Directional breeding and selection for cold hardiness or drought resistance created progeny that had distinct genetic diversity in the tested bermudagrasses. It is evident that an increase in genetic diversity of the existing cultivar pool with the release of one or more experimental selections for commercial use will strengthen and improve bermudagrass systems. Wildlife‐related crimes are the second most prevalent lawbreaking offense globally. This illicit trade encompasses hunting, breeding and trafficking. Besides diminishing many species and their habitats and ecosystems, hindering the economic development of local communities that depend on them, undermining the rule of law and financing terrorism, various cross‐species transmissions (zoonoses) of pathogens, including COVID‐19, can be attributed to wildlife crimes. Wildlife forensics applies interdisciplinary scientific analyses to support law enforcement in investigating wildlife crimes. Its main objectives are to identify the taxonomic species in question, determine if a crime has been committed, link a suspect to the crime and support the conviction and prosecution of the perpetrator. This article reviews wildlife crime and its implications, wildlife forensic science investigation, common forms of wildlife biological evidence, including DNA, wildlife DNA techniques and challenges in wildlife forensic genetics. The article also reviews the contributions of genetic markers such as short tandem repeat (STR) and mitochondrial DNA (mtDNA) markers, which provide the probative genetic data representing the bulk of DNA evidence for solving wildlife crime. This review provides an overview of wildlife DNA databases, which are critical for searching and matching forensic DNA profiles and sequences and establishing how frequent forensic DNA profiles and sequences are in a particular population or geographic region. As such, this review will contain an in‐depth analysis of the current status of wildlife forensic genetics, and it will be of general interest to wildlife and conservation biologists, law enforcement officers, and academics interested in combating crimes against wildlife using animal forensic DNA methods. Tandem repeat genetic profiles used in forensic applications varies between populations. Despite the diversity and security issues in the Sahel that require the identification of victims (soldiers and civilians), Burkina Faso (BF) remains understudied. To fill this information gap, 396 unrelated individuals from BF were genotyped using a MICROREADER 21 ID System kit. All 20 short tandem repeat (STR) loci tested passed the Hardy–Weinberg equilibrium (HWE) test. The combined powers of exclusion for duos (CPE duos) and trios (CPE trios) for the 20 tested loci were 0.9999998 and 0.9999307, respectively. The probability that two individuals would share the same DNA profiles among the BF population was 9.80898 × 10–26. For the X-chromosome STR analysis, 292 individuals were included in this study using a MICROREADER 19X Direct ID System kit. Among the 19 loci, no significant deviations from HWE test were observed in female samples after Bonferroni correction (p < 0.05/19 = 0.0026), except for loci GATA165B12 and DXS7423. The results showed that the combined power of exclusion (CPE) and the combined power of discrimination in females (CPDF) and males (CPDM) were 0.999999760893, 0.999999999992, and 1, respectively. Natural products are a diverse class of compounds with promising biological properties, such as high potency and excellent selectivity. However, they have different structural motifs than typical drug-like compounds, e.g., a wider range of molecular weight, multiple stereocenters and higher fraction of sp3-hybridized carbons. This makes the encoding of natural products via molecular fingerprints difficult, thus restricting their use in cheminformatics studies. To tackle this issue, we explored over 30 years of research to systematically evaluate which molecular fingerprint provides the best performance on the natural product chemical space. We considered 20 molecular fingerprints from four different sources, which we then benchmarked on over 100,000 unique natural products from the COCONUT (COlleCtion of Open Natural prodUcTs) and CMNPD (Comprehensive Marine Natural Products Database) databases. Our analysis focused on the correlation between different fingerprints and their classification performance on 12 bioactivity prediction datasets. Our results show that different encodings can provide fundamentally different views of the natural product chemical space, leading to substantial differences in pairwise similarity and performance. While Extended Connectivity Fingerprints are the de-facto option to encoding drug-like compounds, other fingerprints resulted to match or outperform them for bioactivity prediction of natural products. These results highlight the need to evaluate multiple fingerprinting algorithms for optimal performance and suggest new areas of research. Humans change the environment according to their need, causing modifications and affecting the local biodiversity. This work applied a set of dominant markers to test if urban populations of Cyperus ligularis L. (Cyperaceae) present less genetic diversity compared to forest/non‐urban populations, and if these environmental differences (land use) influence gene flow and population structure. Six available populations of C. ligularis were collected in different types of environments in Recife (Brazil). Overall 15 primers of DNA Amplification Fingerprinting and Inter Simple Sequence Repeat were applied and analyzed by proper statistics. The results revealed moderate genetic diversity, significant pairwise gene flow (Nem), and low differentiation between populations (FST). Land use had no direct correlation with genetic diversity or structure, however, an insular population showed higher differentiation from the remaining. Urban environments may not influence gene flow among C. ligularis populations in such environments; therefore, the genetic diversity is homogeneous in the analyzed area. [ABSTRACT FROM AUTHOR]

DNA barcoding is an essential tool in modern biodiversity sciences. Despite considerable work to barcode the tree of life, many groups, including insects, remain partially or totally unreferenced, preventing barcoding from reaching its full potential. Aquatic insects, especially the three orders Ephemeroptera, Plecoptera, and Trichoptera (EPT), are key freshwater quality indicators worldwide. Among them, Plecoptera (stoneflies), which are among the most sensitive aquatic insects to habitat modification, play a central role in river monitoring surveys. Here, we present an update of the Plecoptera reference database for (meta)barcoding in Switzerland, now covering all 118 species known from this country. Fresh specimens, mostly from rare or localized species, were collected, and 151 new CO1 barcodes were generated. These were merged with the 422 previously published sequences, resulting in a dataset of 573 barcoded specimens. Our CO1 dataset was delimited in 115 CO1 clusters based on a priori morphological identifications, of which 17% are newly reported for Switzerland, and 4% are newly reported globally. Among the 115 CO1 clusters, 85% showed complete congruence with morphology. Distance-based analysis indicated local barcoding gaps in 97% of the CO1 clusters. This study significantly improves the Swiss reference database for stoneflies, enhancing future species identification accuracy and biodiversity monitoring. Additionally, this work reveals cryptic diversity and incongruence between morphology and barcodes, both presenting valuable opportunities for future integrative taxonomic studies. As a follow-up to our teamwork's former work against SARS-CoV-2, eight compounds (ramelteon (68), prilocaine (224), nefiracetam (339), cyclandelate (911), mepivacaine (2325), ropivacaine (2351), tasimelteon (2384), and levobupivacaine (2840)) were revealed as the best potentially active SARS-CoV-2 inhibitors targeting the main protease (PDB ID: 5R84), Mpro. The compounds were named in the midst of 3009 FDA and clinically approved compounds employing a multistaged in silico method. A molecular fingerprints study with GWS, the cocrystallized ligand of the Mpro, indicated the resemblance of 150 candidates. Consequently, a structure similarity experiment disclosed the best twenty-nine analogous. Then, molecular docking studies were done against the Mpro active site and showed the binding of the best compounds. Next, a 3D-pharmacophore study confirmed the obtained results for the eight compounds by exhibiting relative fit values of more than 90% (except for 68, 74%, and 2384, 83%). Levobupivacaine (2840) showed the most accurate docking and pharmacophore scores and was picked for further MD simulations experiments (RMSD, RMSF, Rg, SASA, and H-H bonding) over 100 ns. The MD simulations results revealed the accurate binding as well as the optimum dynamics of the Mpro-levobupivacaine complex. Finally, MM-PBSA studies were conducted and indicated the favorable bonding of the Mpro-levobupivacaine complex with a free energy value of −235 kJ/mol. Massively parallel sequencing (MPS) has emerged as a promising technology for targeting multiple genetic loci simultaneously in forensic genetics. Here, a novel 193-plex panel was designed to target 28 A-STRs, 41 Y-STRs, 21 X-STRs, 3 sex-identified loci, and 100 A-SNPs by employing a single-end 400 bp sequencing strategy on the MGISEQ-2000™ platform. In the present study, a series of validations and sequencing of 1642 population samples were performed to evaluate the overall performance of the MPS-based panel and its practicality in forensic application according to the SWGDAM guidelines. In general, the 193-plex markers in our panel showed good performance in terms of species specificity, stability, and repeatability. Compared to commercial kits, this panel achieved 100% concordance for standard gDNA and 99.87% concordance for 14,560 population genotypes. Moreover, this panel detected 100% of the loci from 0.5 ng of DNA template and all unique alleles at a 1:4 DNA mixture ratio (0.2 ng minor contributor), and the applicability of the proposed approach for tracing and degrading DNA was further supported by case samples. In addition, several forensic parameters of STRs and SNPs were calculated in a population study. High CPE and CPD values greater than 0.9999999 were clearly demonstrated and these results could be useful references for the application of this panel in individual identification and paternity testing. Considering the growing importance of microbiome analyses in forensics for identifying individuals, this study explores the transfer of the skin microbiome onto clothing, its persistence on fabrics over time, and its transferability from the environment and between different garments. Furthermore, this project compares three specific QIAGEN microbiome extraction kits to test their extraction efficiency on fabric samples. Additionally, this study aims to check if these extracts contain human DNA, providing a chance to obtain more information from the same evidence for personal identification. The results obtained show: (1) variations in the skin microbiome between the volunteers, potentially due to their different sex; (2) differences in microbial composition between worn and unworn clothing; (3) the influence of the environment on the microbial signature of unworn clothing; (4) the potential use of certain phyla as biomarkers to differentiate between worn and unworn garments, even over extended periods; (5) a tendency towards extraction biases in the QIAampMP® DNA microbiome kit among the three tested ones; and (6) none of the extraction kits allow for the typing of human genetic profiles suitable for comparison. Genomic data in Gossypium provide numerous data resources for the cotton genomics community. However, to fill the gap between genomic analysis and breeding field work, detecting the featured genomic items of a subset cohort is essential for geneticists. We developed FPFinder v1.0 software to identify a subset of the cohort's fingerprint genomic sites. The FPFinder was developed based on the term frequency–inverse document frequency algorithm. With the short-read sequencing of an elite cotton pedigree, we identified 453 pedigree fingerprint genomic sites and found that these pedigree-featured sites had a role in cotton development. In addition, we applied FPFinder to evaluate the geographical bias of fiber-length-related genomic sites from a modern cotton cohort consisting of 410 accessions. Enriching elite sites in cultivars from the Yangtze River region resulted in the longer fiber length of Yangze River-sourced accessions. Apart from characterizing functional sites, we also identified 12,536 region-specific genomic sites. Combining the transcriptome data of multiple tissues and samples under various abiotic stresses, we found that several region-specific sites contributed to environmental adaptation. In this research, FPFinder revealed the role of the cotton pedigree fingerprint and region-specific sites in cotton development and environmental adaptation, respectively. DNA analysis of traces from commonly found objects like knives, smartphones, tapes and garbage bags related to crime in aquatic environments is challenging for forensic DNA laboratories. The amount of recovered DNA may be affected by the water environment, time in the water, method for recovery, transport and storage routines of the objects before the objects arrive in the laboratory. The present study evaluated the effect of four storage conditions on the DNA retrieved from bloodstains, touch DNA, fingerprints and hairs, initially deposited on knives, smartphones, packing tapes, duct tapes and garbage bags, and submerged in lake water for three time periods. After retrieval, the objects were stored either through air-drying at room temperature, freezing at −30 °C, in nitrogen gas or in lake water. The results showed that the submersion time strongly influenced the amount and degradation of DNA, especially after the longest submersion time (21 days). A significant variation was observed in success for STR profiling, while mtDNA profiling was less affected by the submersion time interval and storage conditions. This study illustrates that retrieval from water as soon as possible and immediate storage through air-drying or freezing before DNA analysis is beneficial for the outcome of DNA profiling in crime scene investigations. Body fluid identification is fundamental in forensic science as it links a specific biological source to a genetic profile, thus providing critical clues for crime scene reconstruction. Blood is one of the most common body fluids found on the crime scene, and several strategies have been developed for its identification in recent decades. Usually, after a preliminary (or presumptive) test to determine the presence of blood (both human and non-human), a confirmatory test is needed to prove that the sample is human blood. Out of the confirmatory tests, immunochromatographic (IC) assays are the most commonly and widely used. This work gives a review of the use of commercial kits specifically developed to detect human hemoglobin or glycophorin A (a surface protein of human red cells) in forensics. Claimed sensitivity varies broadly (ranging from 0.06 to 75 nanoliters of fresh blood), but different values (as low as 0.002 nL) were found during validation procedures. Specificities are high, and the possibility of cross-reaction (with the risk of false-positive results) is so low that it can be considered negligible. False-negative results, however, can be found due to the so-called "hook effect" as well as to the target degradation/modification, which interferes with the Ag-Ab binding. In addition, the chemical compositions of the presumptive test, detergents, and washing can also promote false negative outcomes in peculiar situations. Although IC assays are rapid, inexpensive, specific, and easy to use even on the crime scene, their major limitation is represented by the destructive approach required by this kind of confirmatory test. Since the final goal of the forensic investigation is the genetic typing of a bloodstain, we will describe the strategies developed for IC assays of faint stains as well as the strategies adopted to ensure that exactly the same sample undergoes human blood identification and DNA typing. Cardiovascular disease constitutes a noteworthy public health challenge characterized by a pronounced incidence, frequency, and mortality rate, particularly impacting specific demographic groups, and imposing a substantial burden on the healthcare infrastructure. Certain risk factors, such as age, gender, and smoking, contribute to the prevalence of fatal cardiovascular disease, highlighting the need for targeted interventions. Current challenges in clinical practice involve medication complexities, the lack of a systematic decision-making approach, and prevalent drug therapy problems. Stem cell-derived extracellular vesicles stand as versatile entities with a unique molecular fingerprint, holding significant therapeutic potential across a spectrum of applications, particularly in the realm of cardio-protection. Their lipid, protein, and nucleic acid compositions, coupled with their multifaceted functions, underscore their role as promising mediators in regenerative medicine and pave the way for further exploration of their intricate contributions to cellular physiology and pathology. We present the design and computational examination of a waveguide structure with a parabolic core, implemented in AsSe2 chalcogenide glass. This structure aims to generate a mid‐infrared (mid‐IR) supercontinuum (SC) spectrum, covering the wavelength range of 1.4−7.6 µm. It includes molecular fingerprint region as well as earth's atmospheric transparent window. A pulsed laser providing pulses of peak power of 250 W and pulse width of 200 fs at 2 µm was considered as the pump. The results illustrate that the broadband SC spectrum can be achieved in a short (~8 mm) waveguide pumped with commercially available fiber laser at 2 µm. The novelty of this work lies in the generation of a mid‐IR SC spectrum using a parabolic‐core waveguide pumped in the normal dispersion region, which has not been investigated earlier. The ability to detect low level DNA brings with it the uncertainty of whether the detected DNA is a result of transfer. To address this uncertainty, a simulation study was conducted in which a mock illicit drug packet was placed into the personal bags of individuals. When the average transit time of the packets was increased from around 2 h to more than 14 h, the percentage of the DNA profiles recovered from the packets which could be attributed to the individuals increased greatly from 5.3% to 48.6%. We found that drug packers who were poor shedders could not be included as contributors to the DNA profiles from the drug packets at all and there was a higher chance that individuals other than themselves could be included as contributors to the DNA profile recovered from drug packets. We also found that it was equally likely that the drug packers who had direct contact with the drug packets and bag owners who did not, could be included as contributors to the DNA profiles recovered from the packets. The results in this study highlight the importance of taking into consideration the transit time of drug packet, the shedder status of the alleged packer and the history of an item, when evaluating DNA evidence in the context of illicit drug activities. Despite current advances in body fluid identification, there are few studies evaluating the effect of environmental conditions. The present work assessed the detection of body fluids, blood, semen, and saliva, through lateral flow immunochromatographic (LFI) tests, exposed to tropical weather conditions over time, also evaluating the possibility of obtaining STR (short tandem repeat) profiles and identifying mitochondrial DNA (mtDNA) polymorphisms. Blood, semen, saliva samples, and mixtures of these fluids were deposited on polyester clothes and exposed to open‐air tropical weather conditions for 1 month. The test versions from LFI (SERATEC®, Germany) Lab and crime scene (CS) used for the detection – one per each body fluid type – demonstrated that it is possible to identify body fluids and their mixtures up to 14 days after deposition. At 30 days, blood and semen were detected but not saliva. Full STR profiles were obtained from 14‐day‐old blood samples, and partial profiles were obtained from the remaining samples. It was possible to sequence mtDNA in the samples previously analyzed for STR profiling, and haplogroups could be assigned. In conclusion, this study demonstrated for the first time the possibility of body fluid identification and DNA profiling after exposure to tropical weather conditions for 1 month and also demonstrated the value of mtDNA analysis for compromised biological evidence. The evidence for physical activity (PA) as a major public health preventive approach and a potent medical therapy has increased exponentially in the last decades. The biomolecular mechanisms supporting the associations between PA and/or structured exercise training with health maintenance and disease prevention are not completely characterized. However, increasing evidence pointed out the role of epigenetic modifications in exercise adaptation and health-enhancing PA throughout life, DNA methylation being the most intensely studied epigenetic modification induced by acute and chronic exercise. The current data on the modulation of DNA methylation determined by physically active behavior or exercise interventions points out genes related to energy regulation, mitochondrial function, and biosynthesis, as well as muscle regeneration, calcium signaling pathways, and brain plasticity, all consistent with the known exercise-induced redox signaling and/or reactive oxygen species (ROS) unbalance. Thus, the main focus of this review is to discuss the role of ROS and redox-signaling on DNA methylation profile and its impact on exercise-induced health benefits in humans. The GA118-24B Genetic Analyzer (hereafter, "GA118-24B") is an independently developed capillary electrophoresis instrument. In the present research, we designed a series of validation experiments to test its performance at detecting DNA fragments compared to the Applied Biosystems 3500 Genetic Analyzer (hereafter, "3500"). Three commercially available autosomal short tandem repeat multiplex kits were used in this validation. The results showed that GA118-24B had acceptable spectral calibration for three kits. The results of accuracy and concordance studies were also satisfactory. GA118-24B showed excellent precision, with a standard deviation of less than 0.1 bp. Sensitivity and mixture studies indicated that GA118-24B could detect low-template DNA and complex mixtures as well as the results generated by 3500 in parallel experiments. Based on the experimental results, we set specific analytical and stochastic thresholds. Besides, GA118-24B showed superiority than 3500 within certain size ranges in the resolution study. Instead of conventional commercial multiplex kits, GA118-24B performed stably on a self-developed eight-dye multiplex system, which were not performed on 3500 Genetic Analyzer. We compared our validation results with those of previous research and found our results to be convincing. The diet of fish from Arctic tundra streams was analyzed to compare the efficacy of (1) morphological gut content analysis, (2) DNA metabarcoding, and (3) stable isotope analysis (SIA). Although pairing of morphological and stable isotope methods has become common practice, advances in DNA metabarcoding identification have the potential to improve diet assessment. Thus, we aimed to assess the relative benefits and drawbacks of each technique for the dietary study of freshwater fish. Morphological and DNA methods identified Orthocladiinae and Chironominae as the most frequently consumed taxa. In contrast, stable isotope analysis, which assesses prey assimilation over weeks to months, estimated Arachnida and Tanypodinae as the most significant contributors to diet. The morphological method underestimated the frequency of occurrence of soft-bodied Oligochaeta, while DNA identification underestimated zooplankton frequency of occurrence. Diet estimate differences between methods offered different views of the relative importance of common prey consumed and affected estimates of Levin's Index of niche breadth. Thus, diet analysis methods should be carefully chosen with research questions in mind (i.e., SIA for long-term food web analysis; gut contents to examine feeding on specific taxa) and paired with at least one other method for the most accurate portrayal of diet. Two probabilistic genotyping (PG) programs, STRMix™ and TrueAllele™, were used to assess the strength of the same item of DNA evidence in a federal criminal case, with strikingly different results. For STRMix, the reported likelihood ratio in favor of the non-contributor hypothesis was 24; for TrueAllele it ranged from 1.2 million to 16.7 million, depending on the reference population. This case report seeks to explain why the two programs produced different results and to consider what the difference tells us about the reliability and trustworthiness of these programs. It uses a locus-by-locus breakdown to trace the differing results to subtle differences in modeling parameters and methods, analytic thresholds, and mixture ratios, as well as TrueAllele's use of an ad hoc procedure for assigning LRs at some loci. These findings illustrate the extent to which PG analysis rests on a lattice of contestable assumptions, highlighting the importance of rigorous validation of PG programs using known-source test samples that closely replicate the characteristics of evidentiary samples. The article also points out misleading aspects of the way STRMix and TrueAllele results are routinely presented in reports and testimony and calls for clarification of forensic reporting standards to address those problems.

Background. The development of genetic certificates and nomenclatural standards for newly released cultivars of Malus domestica Borkh. (apple tree) ensures an opportunity to preserve the original and authentic genetic and morphological information on domestic cultivars promising for the south of Russia. The purpose of this study was to develop genetic certificates and nomenclatural standards for 7 apple-tree cultivars developed at the North Caucasian Federal Scientific Center of Horticulture, Viticulture, Wine-making (NCFSCHVW), five of which were released jointly with the All-Russian Research Institute of Fruit Crop Breeding (VNIISPK).Materials and methods. The materials included apple-tree cultivars resistant or immune to scab (Venturia inaequalis (Cooke) G. Winter) from the NCFSCHVW collection stock. Conventional herbarization and DNA fingerprinting methods were used. For cultivar certification, an improved technique of DNA sampling and 12 microsatellite markers were applied.Results. Unique DNA profile certificates and nomenclatural standards were produced for 7 apple-tree cultivars: ‘Vasilisa’, ‘Karmen’, ‘Krasny Yantar’, ‘Soyuz’ and ‘Rassvet’ (with the Rvi6 gene) released jointly by the NCFSCHVW and VNIISPK, plus ‘Zolotoye Letneye’ and ‘Feya’ developed at the NCFSCHVW. The apple-tree herbarium specimens were placed for perpetual storage in the WIR Herbarium of the N.I. Vavilov All-Russian Institute of Plant Genetic Resources (VIR).Conclusion. The results obtained are promising for identification and authenticity verification of new domestic apple-tree cultivars during their reproduction and cultivation.

Commercial panels of microsatellite (STR) loci are focused on the use of DNA of the domestic dog (Canis lupus familiaris) and are often inapplicable for genotyping the DNA of the gray wolf (Canis lupus lupus). We propose a CPlex test system, including one hexa‐ and 12 tetranucleotide autosomal STR loci, as well as two sex loci, that is equally efficient in DNA identification of biological samples of the wolf and the dog. Analysis of molecular variance between samples revealed significant differentiation values (FST = 0.0784, p < 0.001), which allows to use the panel to differentiate wolf and dog samples. Population subdivision coefficients (θ‐values) were calculated for each of the 13 STR loci of the developed test system. It was shown that the values of the genotype frequency for dogs and wolves, without and with considering the θ‐value, differ by three orders of magnitude (for dogs 8.9 × 10−16 and 2.1 × 10−14 and for wolves 1.9 × 10−15 and 4.5 × 10−14, respectively). The use of population subdivision coefficients will allow to identify the most reliable results of an expert identification study and the power of exclusion provided by the STR loci of the CPlex test system makes it possible to achieve a reliable level of evidence in forensic DNA analysis of both wolves and dogs. The test system has been validated for use in forensic identification of the dog and wolf based on biological traces found at crime scenes, as well as for individual identification and establishing biological relationship of animals of these species.
